An electrometric method was used for
measurement of cholinesterase (ChE) activity in the
plasma, brain and liver of 45 days old domestic chickens of both sexes. The ChE assay mixture contained 0.2 ml of plasma or tissue
homogenate, 3 ml of distilled water, 3 ml pH 8.1 phosphate-barbital buffer and 0.12 ml of 7.5% acetylthiocholine
iodide as a substrate. The one-step incubation was for 30 min at 37 ˚C.
Normal reference values for ChE activity (ΔpH/30 min) in the plasma, brain and liver of the
chickens were 0.47, 0.25 and 0.21, respectively. Oral administration of the organophosphorus (dichlorvos, 7
mg/kg) and carbamate (carbaryl,
100 mg/kg) insecticides in chickens induced signs of toxicosis
characteristic of cholinergic overstimulation, and
these signs were associated with significant decreases in plasma (62 and 29%)
and brain (28 and 65%) ChE activities, respectively. The results suggest that the
present electrometric method for measurement of ChE
activity in chickens is efficient in monitoring the enzyme inhibition caused by
organophosphorus and carbamate
insecticides.
